Phenol red is a water-soluble dye used as a pH indicator, changing from yellow to red over pH 6.6 to 8.0, and then turning a bright pink color above pH 8.1. As such, phenol red can be used as a pH indicator dye in various medical and cell biology tests.

Solubility

Practically insoluble in chloroform, ether. Soluble in aqueous alkali hydroxides or carbonates with red color (removed by boiling with zinc dust).

  • Solubility in water: approximately 1g/1300ml.
  • Solubility in alcohol: approximately 1g/350ml
  • Storage and Shelf Life : Store between 10- 30°C. Use before expiry date on the label.

Application

Chemicals and substances that impart color including soluble dyes and insoluble pigments. They are used in INKS; PAINTS; and as INDICATORS AND REAGENTS.Substances used for the detection, identification, analysis, etc. of chemical, biological, or pathologic processes or conditions. Indicators are substances that change in physical appearance, e.g., color, at or approaching the endpoint of a chemical titration, e.g., on the passage between acidity and alkalinity. Reagents are substances used for the detection or determination of another substance by chemical or microscopical means, especially analysis. Types of reagents are precipitants, solvents, oxidizers, reducers, fluxes, and colorimetric reagents.

Water soluble pH indicator used in the 6.8 (yellow) to 8.2 (red) range. Commonly employed as a cell tissue culture pH marker. Under normal conditions, phenol red added to the growth medium will have a pink-red color. Dying cells or overgrowth of contaminants will cause a change in pH resulting in a change in indicator color.

Solubility

Soluble in water yielding a dark red solution (25 mg/ml), alcohol (4 mg/ml), methanol, ethanol (200 mg/ml), and aqueous alkali hydroxides or carbonates with formation of a red colored solution.

  • Storage :Store at room temperature
  • pK: 7.9

Applications

It is commonly employed in cell biology laboratories as a cell tissue culture pH marker. Under normal conditions, phenol red added to the growth medium will have a pink-red color. Usually 15 mg/l is used to produce this effect. If problems arise, such as dying cells or overgrowth of contaminants, this will cause a change in pH resulting in a change in indicator color. This color change is an indication that the medium needs to be replaced. Additionally, Phenol Red could potentially be useful as a differentiation factor.Phenol red in tissue culture media can act as a weak estrogen, especially with human breast cancer cells. Phenol red can be used to measure hydrogen peroxide in supernatants from cultured macrophages in multiwell plates.Phenol Red sodium salt commonly employed in cell biology laboratories as a cell tissue culture pH marker.
